The book begins with a chapter on the function of the epigenome, describing the different ways that epigenetic marks are established and erased, and how those changes affect the structure of DNA and the regulation of gene expression. The discussion is kept brief and a self-contained treatment of the basics of epigenetics is given. This is followed by a chapter on epigenetic gene silencing that includes a short section on the structure and function of repressive complexes, a review of the biochemical and molecular details of DNA methylation and histone deacetylation and a detailed comparison of the biochemical activities of the Su(var)3--9, Trithorax and Polycomb proteins. Subsequent chapters deal with a range of biological processes including stem cell development, sex determination, and the control of immunity and development. The final chapters provide a survey of human diseases associated with epigenetic abnormalities.
After a brief introduction to the function and regulation of regulatory RNAs, the focus of the next two chapters shifts to the role of post-transcriptional gene regulation in developing an immune system, in particular the role of microRNAs in hematopoietic stem cell homeostasis. The final chapter of the book examines the role of regulatory RNAs in cancer development, with a focus on microRNAs, long non-coding RNAs, and circular RNAs. Each chapter ends with a review and an extensive list of references to papers that are not included in the text. The list of references includes recent papers that summarize the state of the field in the past few years.
